so the second method which i'll telling you about for ABO RH blood grouping is by tube method tube method is better than
the conventional slide into it because in the tube method we are doing both the forward
grouping a s well as the reverse grouping so both the grouping by forward
reverse are cross check so chances of HR minimum and that's why it is
called as a full standard for doing them ABO RH blood group so for this procedure
we required atleast 7 tubes which we have to label with the donor ID or with the
patient name the first tube we have to label as Anti-A
second tube Anti-B
Third tube Anti-AB
fourth tube Anti-D
and to do the reverse grouping we have another three tubes
with the first tube with the donor id or with the patient name and ID
We have to write A cell
B cell
and o cell
apart from this we need
Anti sera
Anti sera A
Anti sera B
Anti sera D
Anti sera AB
we also need
the cell suspension
which is should three to five percent of the cells suspension once we are separated the whole blood
into the cell and plasma
Plasma
of the donor or the patient
and commercially prepaid or in the blood bank prepaid
A cell
B cell
and
O cell
so to start with in the first tube will put two drops of Antisera-A
two drops
of Antisera B
two drops of Antisera
AB
and two drops
of Antisera D
after this
will be adding
one drop each of cell suspension
which is three to five percent prepare
one drop in Anti-A
one drop in Anti-B
one drop in Anti-AB
and one drop in Anti-D
to do the reverse typing
first we have to add
two drops
of plasma
in A cell
B cell
and
O cell
after that
commercially prepaid or in the blood bank prepaid
A cell
in the a cell one drop
B cell
one drop in B cell
and
O cell
one drop
after this with gentle shaking we will keep them in the centrifuge
one minute
at 1000rpm
other after gentrification for one minute
at 1000rpm
we will see
that
there is a button shape O+ agglutination
in
Antisera A tube
in the second tube of Antisera B again there is a button shape O+ agglutination
in the third tube again Antisera AB
their is a O+ agglutination
and
in the fourth tube also
of Anti-D there is
a button shape O+ agglutination of Anti-D
so by over the forward typing it is Ab positive
now we have to do the cross check with the reverse type
in the a cell
there is no agglutination
in the B cell
there is no agglutination
and
in the O cell also
there is no agglutination
so
as
it is giving agglutination at O+ reaction in Anti-A Anti-B Anti-AB and Anti-D and in reverse
it is not giving reaction in any of the A cell B cell O cell so again the blood group of the patient
is AB positive
