Tanner Chidester here with my brother who passed-by the lake today, we got 3 sets of
10, we're gonna start on that, I actually took about a half hour to just to get my car
out of the snow, I'm really sick of Utah, so I'm actually might be out of here in about
two months.
Got out of the snow, up here, about to warm up and then we'll show you some of the workout
as usual.
Update on prep, it's going okay.
I still am on my lower calories I just had a little too much food while I was in Texas
and I think I'm just holding some water, I've been a little stressed out lately, but I'll
be fine, talked to my coach, everything is good.
One of the biggest things on prep is not stressing out, in here, gotta hit the workout and then
my brother will start filming.

got back from the gym, just had a cup of oatmeal and then I had about, what was it, a lot of
it, a lot of chicken.
I think I had about, I didn't even measure it - well, I measured it out, it's about 500
grams total and I then I ate about half of it so 500 grams is about 125 grams of protein,
so I got about 50-60 somewhere in there and now I'm gonna get to work.
What I do, is this is where I kinda do my stuff.
I got my new MacBook yesterday and $3,000 for it, but it's a beast so I just sit here
and then I start working and get comfy, do my work.
Get me a little something to eat for I have to go, got some vegetables, I kinda let them
burn a little bit 'cause they're frozen, I like it like that, made a ton of chicken,
some spices, this is a little too much on the cut, it's about 200 grams and now I'm
gonna take off.
Something that had just happened is I got offered a photo-shoot for 8 to 10 grand, for
partial nudity, or full-on nudity, right?
Thing is, it's easy a lot of times when you�re in a tough situation or you, you're like,
"Dang, that's quick cash," to take that but I promise you in the long run, it's just gonna
hurt you.
Stuff like that will never turn out well if you're trying to have some type of business
or career.
So, the only reason I bring that up is don't take the easy way out, wait your time, it
gets tough even in my next YouTube video it's a little bit of a rant, I'm not gonna lie,
in the long run, doing the right things will always pay off over taking the easy way out.

that reason because at the end of the day it hurts your brain.
And I think, as far as you've done the best over the long haul and from the beginning,
a brain himself is someone maybe Steve Cook.
He's never taken any kind of sexy or risqu� photos, you type his name on Google, if you
wanna see, you try to find him, you will see him.
It's not that there's really anything wrong with it, if that's what you want to do, but
at the end of the day, if you're trying to build a brain and stuff, you got to go more
conservative.
It doesn't look as good to most people that's how- what's your goal is.
And if you're trying to build something bigger, you want to put yourself in a good light.
That's all I got guys, I'm gonna go inside, I'm only got- have about 100 grams of chicken,
pretty much maxed out my calories, I'm gonna do a weigh-in on Sunday- On Monday, and I've
got an upper body workout tomorrow.

during the 2017 People's Choice Awards. They also took home the award for Favorite
Group.

Youth and Their Initiatives to Prevent Corruption (Part 2) - Duration: 4:31.
In Indonesia, just like what Miss Nurin said previously, our GPA about the awareness of anti-corruption
it is still very low
and corruption is considered as a high-level crime, together with terrorism and drugs
So what is main reason behind this low awareness of anti-corruption?
Is it related to the quality of our human resources? Is it about the system in our government? Or anything else?
As for the main cause, it is true about what you said previously. There are two factors:
(the quality of) human resources and the system (of the government)
As for the human resource, FLAC try to encourage people to leave this culture of corruption
meanwhile, they are fully aware if corruption will not do any good to them.
But the thing is, if something has been implanted for a very long time, and people then get accustomed to do it
they will no longer feel guilty to do the exact same thing in the future
or, this people might completely have no idea if their actions are categorized as a corruption.
Therefore, that's the first main cause, about the human resources.
Second, it's about the system. In the traffic section, for instance. If we want to make a driving a license
if we don't use money (to bribe the officer) it will be hard for us to get the driving license punctually.
It shows off a system that has been applied there. Who made that system? People themselves for sure.
Therefore, FLAC will always try to build a good character and encourage them to be an anti-corruption.
Everybody can create their own system. And as for characteristic, everybody has a different form of it.
The meaning is, no matter how good the system is, if our human resources still think of doing corruption
then the system will be completely pointless.
So, what are we suppose to do to make sure that our people's characteristics are free from corruption?
Actually, there are some ways to keep our human resources away from corruption
First, FLAC will always ask for influence from family member, to help injecting the value of anti-corruption
to all children.
So it has to be strengthened, start from our own family.
Second, another environment, such as school, will also take a part.
What I said before about 'Jujur Juara' from FLAC, this shows if school can have a huge impact
in spreading the value of anti-corruption to the students
The third is, how do we keep maintaining all of them? By creating networks, such as FLAC
we have to make many small groups that consist of youth with integrity
So in the end, if some Indonesian youth are awarded with certain position in the government
they will remember about the value of anti-corruption that once they declared so loudly
and there will always be friends to help maintaining the value.
It requires many people to help maintaining it.
Couldn't agree more. Because the intention of doing corruption, it will depend on everyone's characteristic
and in order to maintain the value, we have to be surrounded by people with the same intention with us.

Typically, buttock augmentation asymmetries are better corrected with fat
because it is more versatile and can fix a spectrum of different deformities.
Typically, projection-related asymmetries can be fixed by placing a larger implant on one side than the other.
Up to a noticeable difference of 100cc can be corrected.
However, using implants of different sizes to correct asymmetries can be complex.
Placing an implant to correct the defects in the central part of the buttock will affect the surrounding areas.
As a matter of fact, this can even make the asymmetries more prominent
by making the peripheral part of the buttock look more indented.
For this particular reason, a combination of fat transfers is always advised to correct any asymmetries.
The implant will provide the foundation for the projection, while the fat will provide the means
to make the buttocks look more natural and beautiful.
Buttock implants can be used to correct asymmetries
but is better to combine fat and implants for better results.

Proper PCR Cleanup before Sanger Sequencing - Seq It Out #12 - Duration: 3:09.
Why is a clean template for your Sanger sequencing reaction important?
And what are the ways to get it without losing too much precious samples?
Let's find out today.
The sequencing workflow usually starts with PCR also known as Polymerase Chain Reaction,
where the target fragment is replicated into thousands of copies under a controlled condition
using primers, DNA polymerases and deoxynucleotides.
Before the PCR product is ready for Sanger sequencing, you must do one more thing, a
PCR clean up step.
Because Sanger sequencing is a highly accurate technique for you to read DNA sequence base
by base, it is very important to clean up your reaction mixtures so that those unincorporated
primers and dNTPs won't interfere with your results.
So how do you perform this PCR clean up step and what are your options?
There are several methods for PCR cleanup such as ethanol precipitation, bead or column
based purification, and Enzymatic approaches.
Ethanol precipitation is most cost effective but is also the most labor intensive.
Bead or column based purification can be an effective method for many applications.
However, this route tends to be a bit pricier compared to other options.
This method of purification also requires transferring samples in and out of the column
which can often result in the loss of some of your precious samples so this may not be
the best method for those with limited starting materials.
An enzymatic cleanup approach has a really simple and straightforward workflow.
In fact, it's only one single pipetting step!
It is also relatively affordable compared to the bead or column based purification methods.
Basically, you add an enzyme mix to your finished PCR reaction and let it sit at a certain temperature.
But to understand this method better, let's take a look at our lab book.
One of the enzymes, exonuclease I(one), digests excess primers.
At the same time the other enzyme in the mix, alkaline phosphatase, dephosphorylates nucleotides
in the reaction.
Dephosphorylation means that the enzyme renders the dNTPs from your reaction useless, so that
they don't disturb the sequencing reaction.
This is important because the sequencing reaction has a very specific ratio for its nucleotides
and you don't want to change that ratio.
Now the enzymes have done their work and you don't need them anymore.
You can simply heat up the reaction mix to 80 degrees and this will deactivate the enzymes.
The result is a very clean amplicon that is now ready to go into your sequencing reaction.
You don't need to perform any other steps.
And you didn't lose any of your template either.
You are done!
